色譜柱的作用是什么

色譜柱可分為填充柱和開管柱兩大類。多為金屬或玻璃制作。有直管形、盤管形、U形管等形狀。液相色譜通常均采用填充柱。

色譜柱的分離效果取決于所選擇的固定相，以及色譜柱的制備和操作條件。色譜是一種分離分析手段，分離是核心，因此擔負分離作用的色譜柱是色譜系統(tǒng)的心臟。

色譜柱由柱管、壓帽、卡套（密封環(huán)）、篩板（濾片）、接頭、螺絲等組成。柱管多用不銹鋼制成，壓力不高于70 kg/cm2 時，也可采用厚壁玻璃或石英管，管nei壁要求有很高的光潔度。

為提G柱效，減小管壁效應，不銹鋼柱nei壁多經(jīng)過拋光。也有人在不銹鋼柱nei壁涂敷氟塑料以提高nei壁的光潔度，其效果與拋光相同。

還有使用熔融硅或玻璃襯里的，用于細管柱。色譜柱liang端的柱接頭nei裝有篩板，是燒結不銹鋼或鈦合金，孔徑0.2~20μm(5~10&μm)，取決于填料粒度，目的是防止填料漏出。

色譜柱按用途可分為分析型和制備型兩類，尺寸規(guī)格也不同：

1、常規(guī)分析柱（常量柱），內(nèi)徑2~5mm（常用4.6mm，國內(nèi)有4mm和5mm），柱長10~30cm；

2、窄徑柱（narrow bore，又稱細管徑柱、半微柱semi-microcolumn），內(nèi)徑1~2mm，柱長10~20cm；

3、毛細管柱（又稱微柱microcolumn），內(nèi)徑0.2~0.5mm；

4、半制備柱，內(nèi)徑>5mm；

5、實驗室制備柱，內(nèi)徑20~40mm，柱長10~30cm；

6、生產(chǎn)制備柱內(nèi)徑可達幾十厘米。柱內(nèi)徑一般是根據(jù)柱長、填料粒徑和折合流速來確定，目的是為了避免管壁效應。

Chromatographic columns can be divided into two categories:packed column and open column.Most are made of metal or glass.There are straight tube, coiltube, U tube and other shapes.Liquid chromatography usually uses packed columns.

The separation iffect of the column depends on the stationary phase selected,the preparation and operating conditions of the column.Cheromatography is a means of separation and analysis,separation is the core,so the separation column is the heart of the chromatography system.

Chromatographic column is composed of column tube,pressure cap, clamp (seal ring),sieve plate (filter),joint,screw and so on.The column tube is made of stainless steel.When the pressure is not higher than 70kg/cm²,the thick wall glass or quartz tube can also be used.The nel wall of the tube requires high finish.

In order to improve the g-column efficiency and reduce the tube wall effect,the nel wall of stainless steel column is mostly polished. Ther are also people in the stainless steel column NEI wall coated with air plast to improve the finish of the NEI wall,the effect is the same as polishing.

Others are lined with molten silicon or glass for thin pipe strings.The column joint NEI at the liang end of the column is fitted with a sieve plate, which is sintered stqainless steel of titanium alloy.The aperture is 0.2-20μm(5~10&μm),depending on the size of the filler,to prevent leakage of the filler.
